Workshop on High-Throughput Technology

This is a full-day workshop that runs from 9 a.m. to 4 p.m. and includes a working lunch. The focus will be on bringing people up to date on recent developments and applications, integration across technologies, and potential regulatory implications. Forty to 45 minutes will be dedicated to each topic. The speakers will be asked to present 25- to 30-minute talks in order to have 10- to 15-minute discussions after each presentation. Speakers will present available tools/technologies and some applications. The tentative schedule for the workshop is presented below.

Report

The 37th Annual Meeting of the Environmental Mutagen Society program consisted of symposium talks on basic and clinical research on environmental mutagens as well as poster and platform sessions and special sessions designed to facilitate student/postdoctoral presentations and interactions. The meeting is a blend of submitted presentations and other special events, such as invited speaker plenary sessions and education courses, designed to inform attendees about the latest advances in environmental mutagen research. The Environmental Mutagen Society Annual Meeting provided a unique opportunity for translation of basic scientific findings on environmental mutagens because of its multidisciplinary membership with clinical, basic research, and information scientists as attendees.

The Rare Diseases Symposium, titled "Rare Diseases Resulting from DNA Repair Defects," was funded by the generous contribution of the Office of Rare Diseases and was well attended by this challenging group of interdisciplinary scientists. This Symposium was presented at the recent EMS meeting in Vancouver on Sunday, September 17, 2006. The Chairs were P.J. Brooks, Ph.D. NIAAA, NIH, and Kenneth Kraemer, M.D., NCI, NIH.

The first speaker, Dr. Brooks, gave a brief overview of the 19 human diseases currently known to result from mutations in DNA repair genes and pointed out that recurrent themes among multiple DNA repair diseases include increased cancer susceptibility, craniofacial-developmental disorders, and neurological disorders. He then discussed his own work, presenting evidence that a specific type of oxidative DNA lesion called a cyclopurine is the best candidate for the DNA lesion that causes neurodegeneration in xeroderma pigmentosum (XP). His presentation also included new data on the effects of a cyclopurine lesion on transcription by mammalian RNA polymerase II.

The second speaker, Kenneth Kraemer, M.D., discussed the genotype-phenotype relations in the XPB, XPD, and XPG genes, including new data on the phenotypic and clinical manifestations of several novel XPB mutations. He also discussed how different mutations in a single gene, XPD, can give rise to at least three different diseases, XP, Cockayne syndrome, and trichothiodystrophy.

Dr. Kraemer was followed by Dr. Heidrun Intherthal (University of Washington). Dr. Intherthal described her work on the TDP1 enzyme. Mutations in the TDP1 gene are responsible for spinocerebellar ataxia with axonal neuropathy (SCAN1). She presented her hypothesis that the disease results not from a null allele, but a partially functional protein that can become trapped on DNA.

After the break, the fourth speaker, Dr. Sharon Cantor (University of Massachusetts Medical School) described her work on the BACH (BRCA1-Associated C-terminal helicase) protein. Dr. Cantor and others have found the mutations in the BACH1 gene lead to Fanconi anemia, and that BACH is the FANC-J protein. Dr. Cantor also presented data showing that some of the mismatch repair proteins (PMS2 and MLH1) are associated with FANC-J/BACH in human cells. Mutations in PMS2 and MLH1 are associated with another hereditary disease, hereditary non-polyposis colon cancer (HNPCC).

Finally, the last speaker, Dr. Tanya Paull (University of Texas at Austin) focused on the mechanism of the DNA double-strand breaks (DSB) response, including the mechanism of activation of the ataxia-telangiectasia mutated (ATM) kinase. Specifically, she presented data on the important roles of the mediator proteins MDC1 and 53BP1, acting in concert with ATM, in the activation and amplification of the DSB response. The Symposium was very well attended, and numerous questions were asked of each of the speakers. All of the speakers felt that this was a very useful Symposium.

The abstracts from this symposium were published in the Environmental Mutagen Society Journal Environmental and Molecular Mutagenesis, Volume 47, Number 6. This issue was published in July 2006.
